ABSTRACT

OBJECTIVE: To summarize and analyze the diagnostic ideas of new occupational lung tumors in Guangdong Province.METHODS: According to the Law of the People′s Republic of China on the Prevention and Control of Occupational Disease and the GBZ 94-2002 Diagnostic Criteria of Occupational Cancer, the key diagnostic points of 6 new occupational lung tumors diagnosed in Guangdong Province from 2010 to 2011 were analyzed. RESULTS: There were 9 cases of 6 new kinds of new occupational tumors were diagnosed in Guangdong Province in 2010-2011. The cases included 3 occupational lung cancer of coke oven workers, 2 occupational lung cancer caused by asbestos, 1 occupational mesothelioma caused by asbestos, 1 occupational lung cancer caused by arsenate, 1 occupational lung cancer caused by chromate salt, and 1 occupational lung cancer caused by asphalt. During the process, the diagnosis was based on the principles of the comprehensive analysis and the attribution diagnosis, combined with occupational history, occupational disease hazard exposure history, clinical data and auxiliary examination results. If the patients were diagnosed with a primary tumor, the patients′ exposure history to occupational carcinogens should be tracked, traced and confirmed, and the diagnosis should be confirmed by referring to the list of occupational carcinogens and literature reports of the International Labor Organization, and not limited to only the personnel in a particular industry. CONCLUSION: During the diagnostic process of occupational tumors, attention should be paid to confirm the exposure history of occupational carcinogen. The key is to determine the exposure of corresponding occupational carcinogen, the route and the time of exposure and the incubation period.

ABSTRACT

OBJECTIVE: To explore the intervening effects of bone marrow-derived mesenchymal stem cells(BMMSCs) for pulmonary fibrosis of rats exposed to silica dust at different stages. METHODS: Specific pathogen free SD rats were randomly divided into model group,2-week group,4-week group and control group with 6 rats in each group(half males and half females). Rats of the first three groups were one-time endotracheally injected with 0. 5 mL aseptic silica suspension at 30 g/L mass concentration. Rats of control group were injected with 0. 5 mL 0. 90% sodium chloride solution. Rats of 2-week group and 4-week group were injected with 0. 5 mL BMMSCs suspension with cell density was 5 × 10~9/L at 2 weeks and 4 weeks respectively after silica dust exposure,while model group and control group were injected with aseptic 0. 90% sodium chloride solution in the same volume. After that all rats were examined by lung computed tomography(CT) scan,pathological sections were observed,lung coefficient were measured,lung tissue hydroxyproline(HYP) content and serum transforming growth factor β1(TGF-β1) concentration were investigated at the 12 th week after silica dust exposure. RESULTS: Lung CT image showed clean lung field and clear pulmonary parenchyma in control group.Multiple and diffused high density granular shadows of different size and streak/reticular fiber shadows in model group;diffused distribution of very small granular shadows in 2-week group; granular shadows and local reticular fiber shadows in 4-week group,and either the size or the area of granular shadows was smaller than model group. The lung CT value,lung coefficient,lung tissue HYP content and serum TGF-β1 concentration of model group,2-week group and 4-week group were higher than those of control group(P < 0. 05). The lung CT value,lung tissue HYP content and serum TGF-β1 concentration of control group,2-week group,4-week group and model group were elevated in turn(P < 0. 05),while the lung coefficient of model group and 4-week group was higher than that of 2-week group respectively(P < 0. 05).CONCLUSION: BMMSCs could delay pulmonary fibrosis caused by silica dust,and the protective effect is better at early stage than later stage of fibrosis.

ABSTRACT

The aim of this study was to evaluate the expression levels of Cav-1 and MCT4 in the tumor stroma CAFs. Immunohistochemical method was used to detect the expression of Cav-1 and MCT4 in 86 cases of IMPC, with 105 cases of non-specific invasive ductal carcinoma [IDC-NOS] as control. Their differential expression in different histological types were compared and relationship between the expression of Cav-1 and IMPC in the MCT4 and the main pathological features such as age, tumor size and histological grades were analyzed. The study confirmed that the loss expression of Cav-1 on CAFs and the up-regulation of MCT4 may be the possible mechanisms of CAFs in tumorigenesis, which may be one of the molecular mechanisms leading to the poor prognosis of IMPC. The expression of CAFs related proteins in tumor microenvironment suggested that CAFs could be used as the target of tumor therapy and provide new evidence for the treatment of breast cancer

ABSTRACT

Background and purpose:Breast invasive ductal carcinoma (IDC) is the most common breast carci-noma, and the ultrasonographic features of its molecular subtypes has great clinical value. The purpose of this study was to discuss the correlation between ultrasonographic features of IDC and its molecular subtypes.Methods:Ultrasonographic features of 112 patients with preoperative integrated ultrasonographic information and pathologically confirmed IDC from Sep. 2012 to Feb. 2015 were retrospectively analyzed. Based on the immunohistochemistry results of ER (estrogen receptor), PR (progesterone receptor), HER-2 (human epidermal growth factor receptor-2) and Ki-67, these cases were categorized into 4 molecular subtypes: Luminal A group, Luminal B group, ERBB2 positive group and Basal-like group. Results:There were 14 cases (12.5%) in Luminal A group, 62 cases (55.4%) in Luminal B group, 21 cases (18.7%) in ERBB2 positive group and 15 cases (13.4%) in Basal-like group. The 4 molecular subtypes differed in tumor length, lymph node involvement, tumor boundary, tumor shape and internal blood flow on ultrasonography (P0.05).Luminal A group and Luminal B group had less lymph node involvement, more obscure boundary and irregular shape. More internal blood flow, bigger tumor size and lymph node involvement were observed in ERBB2 positive group of this study. Patients in Basal-like group were more likely to have clear tumor boundary, regular tumor shape, bigger tumor size and lymph node involvement on ultrasonogram.Conclusion:Correlation exists between ultrasonographic features and molec-ular subtypes of IDC. This has tremendous clinical significance in the early diagnosis of IDC, preoperative, intraoperative and prognosis evaluation of IDC patients.

ABSTRACT

OBJECTIVE@#To explore the eff ect of pulchinenoside (PULC) on fi broblast-like synoviocytes (FLS) apoptosis in adjuvant arthritis (AA) rats.@*METHODS@#A total of 60 SD rats were randomly divided into 8 groups: A normal control group, an AA group, a low PULC group (50 mg/kg), a middle PULC group (100 mg/kg) or a high PULC group (150 mg/kg) and an ibuprofen (8 mg/kg) group (n=10 per group). FLS from the AA rats was cultured. The expression of Bcl-2, Bax, caspase-3 and the FLS proliferation were detected by the real time qPCR and MTT, respectively. The expression of IL-6 and IL-8 in culture medium was detected by ELISA.@*RESULTS@#Compared with the AA group, the Bcl-2 expression was down-regulated (all P<0.05), the Bax and caspase-3 expression was up-regulated (all P<0.05), and the FLS proliferation was inhibited (all P<0.05). The IL-6 and IL-8 expression was suppressed in the FLS in the PULC groups at different dosages (all P<0.05) as well as in the ibuprofen group (P<0.05).@*CONCLUSION@#PULC may inhibit the FLS proliferation in AA rats by increase in FLS apoptosis.

ABSTRACT

OBJECTIVE:To study the purification technology of total flavonoids from Nelumbinis receptaculum by macropo-rous resin. METHODS:Using adsorption rate and desorption rate of total flavonoids from Nelumbinis receptaculum as index,the type of macroporous resin was selected by static adsorption-desorption tests;With adsorption rate of total flavonoids as index,sin-gle factor test was used to investigate the effects of the concentration of total flavonoids,adsorption time,adsorption speed, drug-loading amount,water amount,volume fraction and amount of eluant and other factors on the purification technology. The op-timal technology was validated. RESULTS:Among 10 kinds of resin,HPD-400 macroporous resin was found to have the best ad-sorption and desorption effects. The optimal purification conditions was as follows as the concentration of total flavonoids 7.00 mg/ml, adsorption time of 3 h,flow rate for sampling of 3 column volume (BV)/h,drug-loading amount of 8 BV,water amount of 6 BV,50% ethanol elution amount of 4 BV. In validation test,mass fraction of total flavonoids from purified Nelumbinis receptacu-lum were 63.88%,62.50% and 63.44%(RSD=1.11%,n=3). CONCLUSIONS:HPD-400 macroporous resin could purify total flavonoids from purified Nelumbinis receptaculum,and established purification technology is stable and practical.

ABSTRACT

Objective To investigate whether the abnormal stress could cause the ultrastructural change of the rats′condylar cartilage in vivo .Methods Fifty‐four SD male rats were randomly divided into condylar neck osteotomy operation(OO)group ,con‐dylar neck fracture operation(FO) group ,and control group ,with 12 rats in each group .Then the OO group rats ,FO group rats and control group rats were executed and sampled at 1st week ,3rd week and 5th week separately to observe the structure of condyle with light microscope(LM ) ,scanning electron microscope (SEM ) and transmission electron microscope (TEM ) .Results LM showed that the general condylar thickness of OO group and FO group were thinner than that of control group at 3rd and 5th week . SEM showed that the collagen and fibre were exposed in the OO group and FO group at 3rd week and 5th week ,the fibre was bro‐ken in the OO group at 5th week .TEM showed that the hypertrophic cell in OO group was more mature than those of FO group and control group at 3rd week .The arrange of fibre was disorder in OO group at 1st week ,the fibre was more disorder and there was even leak in OO group at 5th week .Conclusion The abnormal stress in vivo could result in the ultrastructural change of the rats′condyle ,and this change relates with the stess change degree and affects the growth and development of condyle .

ABSTRACT

<p><b>OBJECTIVE</b>To study the efficacy and safety of thoracoscopic and laparoscopic esophagectomy for esophageal carcinoma.</p><p><b>METHODS</b>Based on hospitalization data from March 2011 to March 2014, 62 cases of esophageal carcinoma treated with thoracoscopic and laparoscopic esophagectomy (minimally invasive group, with 45 male patients) and other 62 cases treated with open esophagectomy (open esophagectomy group, with 45 male patients) were analyzed in a retrospective cohort. The mean age of two groups were (62±9) years and (62±8) years, respectively. Quantitative data were analyzed using t-test, whereas qualitative variables were tested with χ2 test. There were no significant difference in age, tumor location, pathological type, tumor staging between two groups (P>0.05). Perioperative data and three-year clinic outcome were collected. The three-year survival curve were calculated with the Kaplan-Meier method and compared by the log-rank test between the two groups.</p><p><b>RESULTS</b>Compared with open esophagectomy group, minimally invasive group has less amount of bleeding during operations ((231±40) ml vs. (302±37) ml, t=4.63, P=0.000), pleural drainage after operations ((490±41) ml vs. (1 090±43) ml, t=-79.59, P=0.000), and postoperative hospital stay ((16±4) days vs. (17±4) days, t=-2.61, P=0.010). Meanwhile, minimally invasive group has more operation time ((272±39) minutes vs. (242±45) minutes, t=3.97, P=0.000) and total and thoracic cavity retrieved lymph nodes (30±5 vs. 28±4, t=2.39, P=0.018; 15±4 vs. 14±3, t=2.59, P=0.011). Nineteen and 31 patients had postoperative complications and statistical significance difference was found between two groups (χ2=4.83, P=0.028). The three-year survival rate was 73.2% in minimally invasive group and 71.4% in open esophagectomy group. There was no significance difference between two groups (χ2=0.170, P>0.05).</p><p><b>CONCLUSION</b>Thoracoscopic and laparoscopic esophagectomy had the advantages of amount of bleeding, postoperative hospital stay and complications, and had the same three-year survival rate with open esophagectomy.</p>

ABSTRACT

CB2-selective agonists have drawn attention in drug discovery, since CB2 becomes a promising target for the treatment of neuropathic pain without psychoactive or other CNS-related side effects. However, the lack of experimental data of the 3D structures of human cannabinoid receptors hampers the understanding of the binding modes between ligands and CB2 by traditional methods. In the present work, combinational molecular modeling studies including flexible docking, MD simulations and free energy calculations were performed to investigate the interaction modes and mechanism of CB2-unselective agonist CP55940 and CB2-selective agonist GW842166X, separately binding with the homology model of CB2 in a DPPC/TIP3P simulated membrane environment. The binding free energies calculated by MM-PBSA method give an explanation for the activity differences of the studied ligands. Binding free energies decomposition by MM-GBSA method shows that the van der Waals interaction is the dominant driving force during the binding process. Our MD simulations demonstrate that Phe197 could be a critical residue for the binding of CB2-selective agonists. Furthermore, by using the MD simulated binding conformer as a template, the 3D-QSAR studies were performed with the comparative molecular field analysis (CoMFA) approach on a set of GW842166X analogues. A combinational exploration of both CoMFA steric and potential contour maps for CB2 affinities and the MD studied interaction modes sheds light on the structural requirements for CB2 agonists and serves as a basis for the design of novel CB2 agonists.

ABSTRACT

Objective To investigate the prevalence of hyperuricemia (HUA) in the elderly in Tanggu Community of Tianjin,and to analyze the related factors.Methods The elderly inhabitants from one community of Tanggu in Tianjin were randomly selected and their serum uric acid (SUA)levels and metabolic syndrome (MS)-related indexes were examined.There were 622 subjects with complete data (260 men,362 women; mean age 67.4 ± 6.0 years).Results The standardized prevalence of HUA in the elderly in this community was 9.1% in total,and 7.6% in males,10.6% in females,and there was no significant difference between males and females (P=0.281).The levels of SUA were (283.37 ± 78.69) μmol/L in total,(315.80 ± 74.39) μmol/L in males and (260.00 ±73.31) μmol/L in females,and the SUA level was higher in males than in females (P<0.001).The levels of blood urea,creatinine (Cr),triglyceride (TG),body mass index (BMI) were higher in patients with HUA than in patients with normal serum uric acid (P=0.000,0.000,0.002,0.049respectively).The multiple factors regression analysis showed that the SUA level was positively correlated with the levels of blood urea,Cr,TG,low density lipoprotein cholesterol (LDL-C) (P=0.000,0.017,0.000,0.019) and was negatively correlated with the levels of high density lipoprotein cholesterol (HDL-C) and fasting blood glucose (P=0.000,0.001).Conclusions The prevalence of HUA in the elderly is lower in Tanggu Community of Tianjin than in other cities in China.The SUA level of the elderly is significant higher in males than in females.The lower kidney function,hypertriglyceridemia and higher LDL-C are the risk factors for HUA,while elevated HDL-C and fasting blood glucose are associated with the decreased SUA level.

ABSTRACT

Objective To investigate the expressions of estrogen receptor (ER) and progesterone receptor (PR) in different tumor tissues of invasive ductal carcinoma(IDC) of the breast. Methods A total of 32 cases of IDC samples were collected. ER and PR expressions in 4 different sites of every sample were measured by immunohistochemical(IHC) assays. Results Overall, a high concordance in both estrogen receptor and progesterone receptor expressions are observed between different tumor tissue sites. The best Kappa values were 0.789 and 0.810 for ER and PR expression respectively. The worst Kappa values were 0. 563 and 0.672,respectively. Conclusion ER and PR expression is not significantly different in different tumor tissues of the same IDC sample. Repeated testing in different tumor tissue sites has little contributions in clinical settings.

New Chemical Entities (NCEs) development is a systematic long-term project that involves multiple disciplines. The translation research will help to build an advanced R&D system from the basic laboratory research, preclinical studies and clinical evaluation to clinical application of drug, for the purpose of shortening the R&D cycle and accelerate the launch of new drugs. In new drug R&D and its clinical application, drug disposition (absorption, distribution, metabolism, excretion, ADME) properties are important criteria for assessing drug-likeness of candidates. ADME evaluation of NCEs plays an important role in the translation research throughout innovative drug R&D process. Therefore, ADME evaluation at the early stage of drug design and development will be helpful to improve the success rate and reduce costs, and further access to safe, effective drugs.

ABSTRACT

Objective To investigate the prevalence of metabolic syndrome (MS) and its components among the elderly at Tanggu community in Tainjin. Methods The elderly inhabitants were drawn out randomly from one community at Tanggu in Tianjin to examine the MS-related indexes. There were 623 samples with complete data aged (67.4±6.0) years, including 261 males with mean age of (68.3±6.1) years and 362 females with mean age of (66.9±6.0) years. Diagnosis of MS was done by criteria of Chinese Medical Association Diabetes Branch in 2004. Results The standardized prevalence of MS for the elderly in the community was 18.5% with the highest risk group aged 60~69 years. Standardized prevalences of hypertension, high triglycerides, high blood sugar, overweight, obesity, low HDL-C were 49.5%, 38.8%, 25.3%, 42.0%, 4.2%, 3.0% in the elderly. Conclusions The prevalence of MS in the elderly is lower in Tanggu community in Tianjin than in Shanghai, Foshan in Guangdong, Qingdao in Shandong and Beijing city. The condition in female is more severe than in male, the highest prevalence occurs in hypertension, and overweight, high triglycerides and high blood sugar follow it, but few of them are obesity, low HDL-C is fewer too.

ABSTRACT

<p><b>OBJECTIVE</b>To develop a RP-HPLC method for quantitative determination of norisoboldine in Radix Linderae and to provide valuable data for quality control of Radix Linderae.</p><p><b>METHOD</b>The separation was performed on a Phenomenex Gemini C18 column (4.6 mm x 250 mm, 5 microm) at 25 degrees C using a gradient elution of mobile phase A (0.5% formic acid, adjusted pH 2.25 with triethylamine) and mobile phase B (acetonitrile). The detection wavelength was 280 nm.</p><p><b>RESULT</b>The calibration curve showed a good linearity (r = 0.999 9) within test ranges of 0.015-1.509 microg. The average recovery was 99.58% with RSD 1.4%.</p><p><b>CONCLUSION</b>The developed method is simple, accurate and reliable with good repeatability. It is suitable for quality evaluation of Radix Linderae.</p>

ABSTRACT

Objective To summarize the characteristics of blunt renal injury in children and re-port experiences in its diagnosis and treatment. Methods A comparison was done on medical data of 30 children and 36 adults with blunt renal injury. The renal injuries were classified according to the organ inju-ry scaling committee of the American Association for the Surgery of Trauma. Results The rate of severe renal injury in the children group was 50%, significantly higher than 31% in the adult group(P<0.05).Surgical exploration rate in the children group was 40%, lower than 73% in the adult group(P<0.05).In the meantime, the kidney conservation rate of 93% in children group was higher than 64% in adult group(P<0.01). Conclusions The kidney of children is susceptible to severe injury but has sound serf-repairing capability. Therefore, surgical interventions and nephrectomy should be considered cautiously.

ABSTRACT

OBJECTIVE To investigate the ?-lactamase genes in 4 kinds of nonfermenting Gram-negative bacilli isolated from the 98th Hospital of PLA. METHODS Sixty strains of Acinetobacter baumannii,30 strains of Pseudomonas aeruginosa,19 strains of Stenotrophomonas maltophilia,and 15 strains of Flavobacterium were isolated from hospitalized patients.Nine kinds of ?-lactamases genes of TEM,SHV,OXA,CTX-M,PER,VEB,IMP,VIM and GES were analyzed by PCR and verified by DNA sequencing. RESULTS In A.baumannii and(P.aeruginosa),the positive rates of gene of TEM were 100.0% and 66.7%,respectively.SHV gene was positive in 18 of 60 strains of A.baumannii tested,17 of which were SHV-12 subtype ESBLs.The other was a new SHV type ?-lactamase nominated SHV-48.OXA gene was positive in 1 of 30 strains of P.aeruginosa tested,it was an OXA-10 subtype ESBLs.But the rest of genes were all negative. CONCLUSIONS There exist 4 kinds of(?-lactamase) genes at least in nonfermenting Gram-negative bacilli including TEM-1,SHV-12,SHV-48,and(OXA-10.)

ABSTRACT

Objective To identify the factors related to the influence of medical research papers. Methods The influence of 1146 research papers by the hospital in the period 1996-2003 and its relationship with such factors as the backgrounds of the authors and the quality of the papers were studied u-sing regression analysis. Results Nine factors, including the impact factor, the status of the paper (being or not being one based on fund-supported research or representing research achievements), and the status of the author (being or not being chosen for talent cultivation and undertaking or not undertaking research projects), were significantly correlated with the influence of the papers. Conclusion In order to enhance the influence of medical research papers, it is necessary for research' management departments not only to encourage research personnel to publish papers in journals with high impact factors but also to step up efforts to better personnel cultivation, degree education, and project and achievement management so as to set up a quality and distinguished research contingent.
